This innovative and highly sensitive electrophoretic unit was designed by Dr. Narendra P. Singh (Department of Bioengineering, University of Washington) and manufactured by Ellard Instrumentation. This Microgel Electrophoretic Unit is a novel instrument for use in microgel electrophoresis (single cell gel electrophoresis or Comet Assay). One major source of variability is in the electrophoresis step of the assay. This new unit is specifically designed to minimize variability in the results and maintain the sensitivity of the assay.

This new electrophoretic unit has two major innovative modifications that will optimize the sensitive assessment of DNA damage:

  1. The addition of a thick copper plate, (better conductor of electricity than platinum and electrophoretic solution) on either end outside the unit. This copper plate serves as a sink for electricity before its distribution to each slide sitting in the electrophoretic unit. The idea of a copper electric sink is entirely novel.
  2. Individual electrical fields (current supplied from the copper plate) to each slide through its own individual platinum wire.

Conventional electrophoretic units used for comet assay provide a non-uniform electric field. This variability and distortion in the electric field yields a slide location dependent DNA migration pattern from the same sample of cells. Thus, a slide placed in the field of electricity flowing between points of electrode entrance in an electrophoretic solution has more DNA migration than a slide sitting at the other location.

In the MGE unit, the novel copper electric sink and the addition of individual platinum electrodes for each slide eliminates the problem of slide to slide variations in DNA migration patterns, allowing researchers to accurately compare results from different cell samples placed and electrophoresed on slides. This unit is a modification of an existing unit, the TECA 2222 (designed by Dr. Narendra P. Singh, Department of Bioengineering, University of Washington and Dr. Ralph E. Stephens, Department of Pathology, Ohio State University) which has a novel buffer recirculation system (also included in the new unit). These novel modifications make the new Microgel Electrophoretic Unit ideal for sensitive and reliable assessments of DNA damage using the Comet assay and the copper sink and individual electrode features may have applications beyond Comet assay.

TECA (Troy Electrophoretic unit for Comet Assay) is designed (by Dr. Narendra P. Singh, Department of Bioengineering, University of Washington, in Seattle and Dr. Ralph E. Stephens, Department of Pathology, Ohio State University, in Columbus) and manufactured by Ellard Instrumentation. After lysis and proteinase K treatment, slides are placed on horizontal platform of unit. The unit is then filled up with a liter of fluid. To minimize DNA migration patterns from slide to slide, two important features are added to the unit. Both ends of each .010 platinum electrode are connected to power supply (Bk Precision DC power supply Model 1715A. www.bkprecision.com) to provide uniform distribution of current. Similarly for uniform distribution of salts, a pump (from VWR, mini pump II with variable flow, VWR brand Persistalic pump, Speed pump II, medium flow Cat# 57954-016. Telephone: 800-932-5000) is connected to 8 input/output channels for each slide position.
